Created attachment 47 1 image and 3 macros Hi, Sorry for not providing more information earlier. Please run the macros sequentially and compare the intensity measurements. My boss believes that the mean intensity value of the puncta after 16- to 8-bit conversion should be higher than what is shown. She thinks that after conversion, the image may have been inverted, resulting in the low mean intensity value for the 8-bit image. I am not sure if this is really the case but I do not have any intelligent explanation for the observation. Thanks for looking into this. Tuan
